How it works:
The basic principle of the spectrophotometer involves passing light through a sample and detecting the amount of light absorbed at different wavelengths. It works as follows:
Light Source: Emits visible or ultraviolet light.
Slit: Filters and directs light to the sample.
Sample: The sample absorbs part of the incident light.
Monochromator: Separates light into different wavelengths.
Detector: Measures the intensity of light transmitted by the sample at each wavelength.
Computer or Display: Displays the absorption spectrum or concentration of the substance analyzed.
Types of Spectrophotometers:
UV-Vis (Ultraviolet-Visible) Spectrophotometer: Measures light absorption in the UV and visible range. Widely used in the analysis of organic and inorganic compounds.
Infrared Spectrophotometer (IR): Analyzes the absorption of light in the infrared range, being useful in analyzing organic and inorganic compounds.
Fluorimetric Spectrophotometer: Measures the emission of fluorescent light after absorption of light in a sample. Used in fluorescence studies on compounds.
Atomic Absorption Spectrophotometer: Specialized in measuring the absorption of light by atoms in a sample, being useful for the analysis of metals.
Nuclear Magnetic Resonance Spectrophotometer (NMR): Uses the absorption of electromagnetic radiation to study molecular structure.
Mass Spectrophotometer: Measures the mass of ions formed by the ionization of a sample. Used in analysis of organic compounds.
Emission Spectrophotometer: Measures the light emitted by a sample after excitation by radiation. Used in compound emission studies.
